Police allege the woman was stabbed several times during an armed robbery at the A&B Food Store, along the popular Elizabeth Street cafe and restaurant strip, about 12.20pm on Saturday.
Emergency crews worked to save the woman in store and she was taken to Royal Hobart Hospital, but she died a short time later.
Police allege the man inflicted a flurry of stab wounds to the woman’s upper body before fleeing the Elizabeth Street scene with the alleged murder weapon and a till cash drawer.
The man was arrested at a Claremont property about 11pm on Saturday, police said.
He was interviewed by detectives at a Hobart police station for a number of hours before being charged in the early hours of Sunday.
The man appeared in the Hobart Magistrates Court on Sunday.
