The victory saw the Roosters regain the Nathan Duncan Memorial Trophy.
Playing at Weeraman Field for the first time this year, Muswellbrook started well and surprised its more-fancied neighbours to hold a six-point lead at the first change.
Singleton was able to gradually gain the ascendancy in the second quarter before running out 39-point winners.
The Cats continue to show improvement in their first season in the Black Diamond Plate competition.
And, after playing four of the more fancied teams in the opening rounds, will relish the opportunity to challenge the other sides in coming weeks with a win not far away.
Kyle Bray, Nigel Smith, Dominic White and Adam Higham were the best for the Cats with Mathew Merrett lively up forward finishing with two goals.
GOALS: Mathew Merrett 2, Lindsay Paterson 1, Adam Higham 1, Lincoln Churcher 1, Cameron Benkovic 1
BEST: Kyle Bray, Nigel Smith, Dominic White, Adam Higham, Mathew Merrett, Riley Warner
Muswellbrook’s women’s team also went down to Singleton by 63 points, however put in a gallant performance with limited numbers.
Danielle Nicol, Natalie Heard, Demi Frazer and Ashleigh Barrowcliffe were their best players.
Next week, Muswellbrook is on the road when its takes on Newcastle City at No 1 Sportsground.
