Shortly after 3.30pm, the Westpac Rescue Helicopter Service (WRHS) dispatched a couple of choppers to the scene, near Wambo Street.
Traffic was affected in both directions as emergency services, a Crash Investigation Unit and council staff attended the incident.
According to a WRHS media spokesperson, a motorbike with a pillion passenger collided with the rear of a trailer, being towed by another vehicle.
The rider and their companion suffered severe injuries.
The duo, a 50-year-old man and a 55-year-old woman, was then treated and stabilised by NSW Ambulance paramedics and the WRHS critical care team.
Both patients were assessed and flown to John Hunter Hospital in a serious condition.
After arriving at the scene, shortly following the first aircraft, the second helicopter returned to the WRHS base.
