Police said the incident was domestic-related and have laid charges against a 22-year-old man.
A Westpac Rescue Helicopter Service spokesman said the chopper flew the 47-year-old man to the John Hunter Hospital after they were called into action shortly after 1am on Friday.
Paramedics had arrived on the scene and begun treating the man before he was driven to Muswellbrook Hospital's helipad to meet the helicopter and its critical care medical team.
The man received further treatment to stabilise him before he was flown to Newcastle in a serious but stable condition.
Hunter Valley police said a 22-year-old man and 17-year-old girl were at a Wollombi Road home about 12.30am on Friday when a 48-year-old man and another teenager attended the home.
"Police have been told the 48-year-old man became aggressive towards the 22-year-old man before there was a struggle between the pair and the older man was stabbed in the shoulder and abdomen," police said in a statement.
Investigators established a crime scene, which has been combed by specialist officers.
Police said the younger man suffered minor injuries and was taken to Muswellbrook hospital.
After his release he was taken to Muswellbrook Police Station and charged with contravene prohibition/restriction in AVO and breach of bail.
The 22-year-old was refused bail to face Muswellbrook Local Court.
Investigations are ongoing, police said.
