With previous co-leaders Denman Devils suffering a shock loss to defending premiers Scone Thoroughbreds on Friday, the Rams sit atop of the ladder following a hard-fought 30-22 win.
In an entertaining, and somewhat fiery, opening half, Muswellbrook enjoyed a 24-12 lead at the break.
Ironically, it was a try to former Greyhound Jake Hardy, which gave the Rams a handy advantage.
Singleton refused to surrender in the second period – and actually outscored Muswellbrook 10-6.
Meanwhile, the Aberdeen Tigers secured their maiden triumph for 2017, beating the Greta Branxton Colts 38-32 at Greta.
In round four this weekend, the Greyhounds face the Colts at Pirtek Park on Saturday.
On Sunday, the Rams host the Devils at Olympic Park while the Tigers travel to Scone to tackle the Thoroughbreds.
