The fourth-placed outfit upstaged the hosts, who occupied third spot on the ladder, 26-4 on the back of an impressive first half.
In fairness to Singleton, the side was missing strike player Holly Fisher-Webster, which affected the home team’s attacking prowess.
The Ramettes surged to a dominant 20-0 lead at the break, courtesy of a terrific double to Harmony Liddell, and four-pointers to Bronte Collins and Cressa Harris.
Shae Pemberton landed two goals.
The Greyhounds responded early in the second stanza, via perennial tryscorer Maddie Sonter, who tried valiantly all match along with Jacki Hicks and Ebony Ford.
But, Muswellbrook hit back almost immediately when Lacey Walton touched down out wide.
Pemberton put the icing on the cake with a successful conversion.
Despite the result, both squads are expected to meet again in the upcoming play-off series, with the likes of Murrurundi Mavettes and Merriwa Magpies.
MUSWELLBROOK RAMETTES 26 (Harmony Liddell 2, Bronte Collins, Cressa Harris, Lacey Walton tries; Shae Pemberton 3 goals) defeated SINGLETON GREYHOUNDS 4 (Maddie Sonter try)
